O.D. Prysyazhna. Infringement of NO-dependent regulation of contractive reactions of heart and vessels at experimental diabetes mellitus. - Manuscript.

The mechanisms of derangement of contractive reactions of heart and vessels et experimental diabetes mellitus type I was a subject of this research. The thorough investigation of disturbances of endothelium-dependent reactions of aorta isolated preparations, portal vein strips and also coronary vessels and heart is spent at streptozotocininduced diabetes. We have revealed a complex of disorders of contractive reactions of myocardium and vascular smooth muscles, which can be caused by chronic hyperglycaemia.

At an animal with experimental diabetes mellitus the depression of endothelium-dependent relaxation of aorta and coronary vascular smooth muscles, infringement of the length - force of contractions ratio and growth of vascular smooth muscles stiffness, increase of oxygen cost of the vascular smooth muscles work is revealed. On Langendorff isolated heart the essentially increase of left ventricular diastolic stiffness was shown at experimental diabetes. These phenomena were accompanied with reduction of Franc-Starling curve phase plateau extension (length - force of contractions ratio) and increase of oxygen cost of the myocardium work.

The basic mechanisms of glucose toxicity are reduction of NO synthesis and development of oxidative stress.

A role of depression of NO synthesis in development of such infringements specifies the revealed disturbances of nitric oxide system functioning et experimental diabetes mellitus. It proves to be true by development of disorders of contractive reactions of heart and vessels similar to diabetic at control animals at blocking of NO-synthase by L-NAME and partial restoration of contractive activity of heart and vessels at rats with experimental diabetes after long reception of the predecessor of NO biosynthesis L-arginin.

About participation of oxidative stress in occurrence of alteration of cardio-vascular reactivity at streptozotocininduced diabetes mellitus testify: increase of the contents of dien conjugates in vessels and heart of diabetical rats; development of changes of contractive reactions of heart and vessels similar to diabetic at control animals under action of oxidative stress inductor tert-buthyl hydroperoxide and partial restoration of the choosen reactions after introduction of antioxidative substance melatonin.

Reduction of MPTP sensitivity to inductors of its opening together with data on myocardial contractility testifies to increase of ischemic tolerance of myocardium at experimental diabetes mellitus.
